The word "guess" is to estimate or conclude (something) without knowing enough to be 100% sure that you are correct.

I suppose so is more so used in a sense when the speaker realises that they might have been wrong and have no been proven wrong. Also when not confident.
I guess so usually used when a disagreement has happened and the speaker doesn't agree but would rather just move on from the conversation.
