This is less intrusive and more polite:
--> "What kind of work do you do?"
This allows the person being asked to be as vague or specific as he wishes. This is important because many people don't like to talk about what they do or who they work for - many times because they're not comfortable giving information that may reveal their wealth, or lack of wealth to others.

The second one is very common but you could also ask:
What company do you work for?
To this they would answer what their company name is, they might also tell you what the company does if it is not a widely known company.
For example they could answer:
“I work for Apple, it’s a phone company.”
Where do you work?
To this they would likely answer what the place is and where it is
For example:
“I work for the clinic on the corner of Maine Street.”
“I work for Walmart that’s just off the freeway.”
What job do you have?
This would probably be replied to with a very simple answer.
For example:
“I work at a coffee shop”
“I am a doctor”
