"Absolutely essential" refers to anyone that is needed to be able to film the scene, such as the actors, directors, cameramen, etc. "Anyone else" refers to literally any other person that is not a part of that list of people. Those other people would distract the people that are needed to film the scene.
Distract: prevent (someone) from giving full attention to something.

"Stressing the fact" means that this person is making this point the most important. He wants you to pay the most attention to this one thing. He wants to "stress the fact" that the movie set is closed, or off limits, to people who aren't needed in it. Therefore, people can't just walk around on the set unless they're a part of the scene.
"Anyone else" are those people who AREN'T necessary for the scene.
If A and B are actors in the scene, but C and D are actors in another scene, then C and D aren't necessary for the current scene, so the director doesn't want them walking around the set.
