@b4briliant: "what are you on?" Can be used a joke to indicate the other person is just bonkers/hyper and you're implying they're on drugs - quite funny actually.
As for the first one, it's just our way of saying. "What are you talking about?"
You might also hear "what you on about" in very informal speech.
They can both give negative indications to the other person depending on your relationship with them and the situation because they can either be taken as negative or for fun.
If I used these with my best friend then they'd definitely just be for fun.
