If you want to say that you have the ability to speak English, then you should say:
I speak English.
For the other sentence that you asked about, we actually wouldn’t use “speak” there. We would say:
I told the story in English.
or
I talked about the story in English.

You can say "I speak in English", but it's probably better to just say "I speak English", which is what most people say.
And for your second question, it depends on the sentence. Could you give some more examples?

"I speak about the story in English?"
for example, from auditorium, from the stage, speaking the story. like TED speaking.
"speak" is used in what case One-side, right.
